Strain Establishment

In the severely immunodeficient mice NPG, mouse Fcgr2b gene and Fcer1g gene-specific sgRNA (single-guide RNA) were used to guide Cas9 nuclease to cut the genome, causing DSBs (Double-Stranded Breaks). The NHEJ (Non-homologous end Joining) repair pathway was used to repair these breaks, resulting in frameshift mutations that achieved the knockout of the Fcgr2b and Fcer1g genes. This strain lacks T, B, and NK cells, and all four Fc gamma receptors (FcgRs) have been knocked out. This strain can more accurately evaluate the efficacy of antibody drugs.

 

Application Fields of NPG-FcgRs Mice

1. Evaluation of the efficacy of antibody drugs. It better simulates the real process of the Fc end of antibody drugs in the human body, promoting the accuracy of preclinical efficacy validation of antibody drugs.
